Abstract

This study aimed to investigate the potential presence of Salmonella sp. in tiger shrimp (Penaeus monodon) and evaluate the microbiological safety of the sample in relation to applicable food safety standards. The analysis involved laboratory preparation, preparation of culture media, sterilization of laboratory equipment and glassware, and microbiological examination using selective media and biochemical identification tests. Sterilization was performed before and after testing to minimize contamination and ensure the reliability of the analytical procedures.

The results revealed that the isolates obtained from tiger shrimp exhibited biochemical characteristics that did not fully correspond to the typical profile of Salmonella sp. Black colonies were observed on Hektoen Enteric Agar (HEA) and Bismuth Sulfite Agar (BSA), suggesting hydrogen sulfide (H₂S) production, whereas white colonies without black centers were observed on Xylose Lysine Deoxycholate (XLD) agar. Biochemical testing revealed positive citrate, urease, and malonate reactions, as well as positive lactose and sucrose fermentation, while indole and Voges–Proskauer (VP) tests yielded negative results. These findings were inconsistent with several characteristic biochemical reactions of Salmonella sp. and suggested the possibility of other members of the family Enterobacteriaceae, particularly Citrobacter spp., which may exhibit similar colony morphology and H₂S production. Therefore, the isolates could not be conclusively identified as Salmonella sp. based on the tests performed. Further confirmatory analyses, including serological testing or molecular identification, are required to establish the identity of the isolates accurately.
